A pre-clinical study for pharmacognostical and chemical standardization of aqueous and alcoholic extracts of Ayurvedic spermatogenetic formulation using spectroscopy & chromatography (including GC-MS) methods

Abstract:
An Ayurvedic formulation prepared from root powder of Sida cordifolia Linn. and Glycyrrhiza glabra Linn. is standardized through pharmacognostical and phytochemical pre-clinical studies for treatment of male sexual disorders. Flavonoids, carbohydrates, glycosides, tannin and Nitrogen were found present in both alcoholic and aqueous extracts. The results also showed high concentrations of flavonoidic compounds (80.31 µg QE/mg) in alcoholic and high phenolic content (28.60 µg GAE/mg) in aqueous extract. HPTLC analysis at 280 and 360 nm indicated presence of Ellagic acid and Gallic acid in both extracts. Similarly, HPLC analysis at 276 nm showed elution of twelve compounds & confirmed presence of Vanillin in both extracts. UV-Visible spectroscopy scanning showed peaks at 822.5, 261 and 194.5 nm in aqueous and at 896, 361, 315, 273.5 and 211 nm in alcoholic extract. FTIR analysis indicated presence of -CH3 out-phase stretching, -NH2 bending, (N)-CH3 in phase bending, C=O in-phase rocking functional groups in aqueous and C=N stretching, –CH2 bending, (N)-CH3 in-phase bending, Aryl-CH3 in-phase bending, C-O-C out-phase stretching and ̶ O-H Wagging functional groups in alcoholic extract suggesting presence of phenolic, flavonidic & alkaloidal compounds possibly responsible for its antioxidant, analgesic, antipyretic, anti-inflammatory and antimicrobial properties.
